In this study, by means of the Taguchi Method-L9 orthogonal array experimental design, the extraction conditions of the UAE method for 4 parameters at 3 levels have been optimized. The use of this method allowed to achieve a high degree of extraction of sugars and sugars alcohols from foliage of conifers. Also, a high degree of extraction was connrmed by the multiple successive extraction method. The hydrophilic interaction chromatography-tandem mass spectrometry method had also been developed and validated for the separation and determination of 17 sugars and sugar alcohols. So, it was shown that using mass spectrometric detection and LC separation amide columns can achieve record low detection limits and excellent analytical characteristics of the method without derivatization. The UAE and HPLC-MS/MS methods were tested on the series of 10 plants (two samples per species representing: Abies, Larix, Picea, Pinus (Pinaceae) and Juniperus (Cupressaceae) covering all genera of the boreal conifer forests (taiga) of Eurasia. Also, the recovery stability was shown for a series of samples by the multiple successive extraction method. These results connrm the developed UAE and chromatographic methods with optimized parameters as universal strategy for sugars and sugars alcohols content determination in conifers.
