Fluorescence Polarization Immunoassay (FPIA) is homogeneous method based on the measurement of fluorescence polarization value for reaction mixture of sample (typically 10-50 µl) and immunoreagents: fluorescent-labelled antigen (tracer) and specific antibodies, and followed measurement of fluorescence polarization of reaction mixture. The total time required for an assay is few seconds for one sample or minutes for up to 96 samples in microtiter plate. The FPIA is a simple, quick, accurate and cost-efficient immunoassay which is readily suitable for quantitative detection of multi analytes. The fluoroquinolones are class of synthetic antibiotics that are widely used for veterinary purposes and must be detected in food products. More than 20 different fluoroquinolones are currently applied in veterinary and must be detected at level of MRL, which varied around 100 ng/mL. The results for development of FPIAs for specific and class-specific detection of fluoroquinolones using different combination of antibodies and tracers will be discussed. Moreover, the results for stereoselectivity of FPIA for Ofloxacin will be presented with stereo-specific immunoreagents because the Ofloxacin is a racemic mixture and its active form is the (−) S isomer – Levofloxacin.
