Целью проекта является выяснение фундаментальных закономерностей накопления биологически-активных веществ разных видов тиса (Taxus spp.) – дитерпеноидов таксанового ряда (таксоидов) – в интактных растениях и инициированных из них культурах клеток. Эта информация необходима для понимания специфики метаболизма и функциональной значимости таксоидов для интактных растений и клеток in vitro.
Plants are one of the most complex living organisms in term of their chemical composition. Secondary metabolites represent the variable part of this composition. New secondary metabolites finding, their biosynthesis and accumulation mechanisms are the most important tasks for modern plant physiology and biochemistry. Given that secondary metabolites determine the physiological activity of most medicinal plants, studies on secondary metabolites also have applied value. The taxane diterpenoids — taxoids — represent a unique group of secondary metabolites. This group is exclusively found in plants of the Taxaceae family (species of the genus Taxus spp. and Austrotaxus spicata). The compounds in this group have a complex chemical structure: more than 500 taxoids of 11 structural types has been described. Moreover, taxoids have pronounced physiological activity. For example, the taxoid paclitaxel is one of the most popular antitumor preparations of plant origin. Together, this causes an increased interest in this group of secondary metabolites. However, the strict endemicity and slow growth of yew make it difficult to study the secondary metabolism of these plants. At the same time, the formation and accumulation of taxoids, a highly diverse chemical structure group, have undoubted fundamental and practical significance. According to some estimates, taxoids make up about 4% of the number of plant-derived diterpenoids currently known. Taxus spp. cell and tissue culture in vitro can be used as an alternative experimental approach to studying the formation and accumulation of taxoids. However, the secondary metabolism in plant cells in vitro and in planta can differ significantly. The mechanisms of this phenomenon are still not entirely clear. The main reasons discussed are: changes in the cell differentiation (in intact plants, secondary synthesis occurs in specialized cells) and the consequences of combining continuous cell proliferation with the secondary metabolites formation/accumulation in in vitro cultures. To correctly interpret the changes in secondary metabolism in plant cells in vitro, it is necessary to compare this process in an intact plant and in a culture of cells obtained directly from this plant. To date, there are no such studies for yew cell cultures. Despite the particular researchers interest in taxoids, their formation and accumulation in vivo and in vitro have been studied very fragmentarily. For example, from the entire structural diversity of taxane diterpenoids, detailed systematic studies (in intact plants and cell cultures) were carried out only for 13-hydroxylated taxoids (such as paclitaxel and its precursors and/or homologs). Meanwhile, to elucidate the general laws of taxoid accumulation, a detailed comparative analysis of namely the different structural groups of these compounds in intact plants (including their organ-specific distribution) and in yew cell cultures is necessary. The present project is dedicated to the study of accumulation of yew species (Taxus spp.) biologically active substances – taxane diterpenoids (taxoids) – in intact plants themselves and in cell cultures initiated from these plants. Such information can help to understand the specifics of taxoids metabolism and taxoid functions in intact plants and cells in vitro. The following experimental tasks are planned to achieve this goal. A detailed analysis of taxoids of different structural groups (13-, 14-hydroxylated and others) in intact plants of several yew species (T. cuspidata, T. wallichiana, T. baccata (different cultivars)) will be carried out. An organ-specific distribution of structurally distinct individual diterpenoids in intact plants of yew species will be performed for the first time. Furthermore, a detailed comparative study of the taxoids composition in Taxus spp intact plants and cell cultures obtained from them will be carried out.
По окончании проекта ожидаются следующие результаты: 1) Будет проведен детальный анализ таксоидов в интактных растениях Taxus spp. (T. cuspidata, T. wallichiana, T. baccata (разные культивары)); 2) Будут выявлены особенности распределения таксоидов разных структурных групп по органам/тканям растений Taxus spp. (T. cuspidata, T. wallichiana, T. baccata (разные культивары)); 3) Будет проведен подробный сравнительный анализ состава таксоидов разных структурных групп в интактных растениях Taxus spp. (T. cuspidata, T. wallichiana, T. baccata (разные культивары)) и инициированных из них каллусных культурах клеток; 4) Будут выявлены закономерности накопления таксоидов разных структурных классов в клетках Taxus spp. in planta и in vitro;
В реализации проекта будут принимать участие сотрудники кафедры физиологии растений МГУ имени М.В.Ломоносова, а также сотрудники филиала данной кафедры в ИФР РАН. В 2007 году в ИФР РАН стартовали работы по созданию коллекции каллусных и суспензионных культур клеток разных видов тиса. В настоящее время в этой коллекции поддерживаются в активно растущем состоянии в общей сложности 23 линии каллусных и 7 линий суспензионных культур клеток тиса. На кафедре физиологии растений МГУ активно проводятся работы по изучению физиологии и биохимии вторичного метаболизма интактных растений и культивируемых in vitro клеток растений. Впервые было проведено сравнительное исследование образования таксоидов разных структурных групп в культурах клеток трех видов тиса – Taxus baccata, T. × media и T. canadensis. С помощью ВЭЖХ-МС и спектроскопии ЯМР установлено, что таксоиды присутствуют во всех исследованных культурах клеток, при этом основными являются 14β-гидроксилированные производные (синенксан C и ряд других таксоидов группы тайванксана); 13α-гидроксилированные таксоиды (десциннамоил таксинин J и таксуспин F) были обнаружены только в культуре клеток T. × media. Таким образом, впервые показано, что образование 14β-гидроксилированных производных такса-4(20),11-диена является специфической особенностью культивируемых in vitro дедифференцированных клеток тиса, отличающей их от клеток интактных растений. Также впервые было показано образование неполярных (полиацилированных) 14-гидроксилированных таксоидов (таксуюннанин C) в культуре клеток T. canadensis. Коллектив заявителя имеет опыт работы в области фитохимического анализа (с применением разных методов и подходов (от классической ТСХ, препаративной хроматографии, расшифровки спектров ЯМР и результатов масс-спектрометрии высокого разрешения до рутинных количественных анализов с применением ВЭЖХ-УФ и ВЭЖХ-МС)) вторичных метаболитов растений изопреноидной природы (тритерпеновые и стероидные гликозиды, фитоэкдистероиды и дитерпеноиды).
Будет проведен подробный сравнительный анализ таксоидов разных структурных групп (не только 13-гидроксилированных, как в большинстве имеющихся сейчас работ, посвященных данной теме) в интактных растениях разных видов тиса, и инициированных из них культурах клеток in vitro. Подобные исследования необходимы для выяснения специфики метаболизма и функциональной значимости таксоидов для интактных растений и клеток in vitro.
