Изучение возможностей неорганических наночастиц в методах люминесцентного определения биологически активных, в том числе лекарственных, веществ
The development of new approaches in the field of personalized medicine and health care is inextricably linked with the development of means and methods of chemical analysis that provide information on the content of medicinal and other biologically active substances in medical samples, pharmaceuticals and other objects. Screening methods are of great importance as far as they allow to conduct a relatively simple and quick preliminary assessment of the content of target substances in the object of analysis in order to decide on the advisability of its further detailed analysis using powerful and expensive methods. The main requirements for preliminary screening methods are high sensitivity, rapid determination, simplicity and mobility of the equipment used, low cost and environmental safety of the reagents. Analytical methods that meet these requirements include luminescence spectroscopy. This method has long been successfully used for the sensitive determination of various biologically active substances. It is advisable to investigate new luminescent reagents and preliminary sample preparation procedures for development of this method. Of particular interest in this regard are nanoobjects, which have been actively studied recently as an alternative to traditional reagents and sorbents. Thanks to their non-classical properties, these objects make possible both improving the characteristics of known approaches and implementation of new analytical procedures. The goal of this project is to study the capabilities of inorganic nanoparticles in methods for the luminescent determination of biologically active substances, including medicinal substances. The project intends to consider inorganic nanoparticles both as direct luminescent reagents and as a tool to simplify sample preparation before analysis. In the first case, silicon nanodots will be studied. These environmentally friendly nanoobjects have their own luminescence, which is easily excited in the visible range of the spectrum. This fact reduces the requirements for the excitation source and the equipment used. Moreover, the presence of a large number of active functional groups on the surface of nanoparticles is favorable from the point of view of interactions with various organic molecules. Preliminary data, in particular, indicate the possibility of luminescent determination of such vital hormones as catecholamines with their help. Another type of promising nanoobjects that is expected to be studied are copper nanoclusters. Compared to the well-known luminescent silver nanoclusters, copper nanoclusters have been studied relatively little. At the same time, they may be of interest due to their excellent spectral characteristics, selectivity, and the significantly cheaper precursor used for their synthesis. Their important feature is increased selectivity for sulfur-containing substances. As part of the project, the capabilities of these nanoobjects will be studied to solve the problems of determining sulfur-containing amino acids. Sample preparation plays a major role in luminescence spectroscopy, allowing both to increase the sensitivity of the analysis due to concentrating procedures and to reduce the interfering influence of the components of the object matrix. The sorption properties of nanoparticles can be effectively used to increase the efficiency of this stage of analysis. This project involves the development of sorption systems based on inorganic nanoparticles for the extraction and concentration of luminescent substances. In order to simplify the sample preparation procedure, we will consider sorbents based on superparamagnetic magnetite nanoparticles, which make possible rapid separation of concentrated analytes using the method of magnetic solid-phase extraction. As part of the project, it is planned to evaluate the effectiveness of these sorbents in relation to the luminescent determination of antibacterial substances of the tetracycline class.
По итогам выполнения проекта будут: – Cинтезированы и охарактеризованы с точки зрения аналитических свойств кремниевые наноточки, модифицированные на поверхности аминогруппами. Кремниевые наноточки представляют из себя экологически дружественные нанообъекты, обладающие выраженными люминесцентными свойствами – применение их для целей химического анализа способно упростить и повысить доступность аналитических процедур при определении биологически важных веществ. – Получены и детально охарактеризованы новые сенсорные аналитические системы на основе нанокластеров меди, детально изучены возможности использования нанокластеров меди для люминесцентного определения таких приоритетных биологически активных соединений, как серосодержащие аминокислоты. Это позволит применить выявленные закономерности для создания новых оптимизированных подходов к решению задач определения лекарственных веществ. – Предложены сорбционные системы на основе суперпарамагнитных наночастиц магнетита для выделения и концентрирования люминесцирующих веществ. Помимо магнитных свойств наночастицы магнетита обладают уникальными сорбционными свойствами, которые обеспечивают селективные взаимодействия с определенными классами веществ, например, с тетрациклинами. В рамках проекта будут систематически исследованы основные факторы, влияющие на сорбцию тетрациклинов, полученные данные будут положены в основу разработки методик люминесцентного определения тетрациклинов после их выделения на магнитных наночастицах из объектов окружающей среды или пищевых продуктов.
Руководитель проекта имеет богатый опыт работы с нанообъектами различной природы (золотые наночастицы, наностержни золота и серебра, люминесцентные нанокластеры золота), а также имеет определённый задел по использованию различных наночастиц в спектрофотометрии и люминесцентной спектроскопии. Другие члены коллектива также в своей работе успешно использовали наночастицы для разработки спектрофотометрических методик определения различных соединений, а также для целей разделения и концентрирования. К настоящему моменту научный задел по теме проекта состоит в том, что были начаты исследования о возможности использования неорганических наночастиц для люминесцентного определения различных соединений: научным коллективом предложены способы определения аденозинтрифосфата и глутатиона с использованием различных систем на основе неорганических наночастиц. Оптимизирована методика получения суперпарамагнитных наночастиц магнетита, у исполнителей имеется опыт работы в области магнитной твердофазной экстракции с полимерными магнитными сорбентами. Получены первые положительные результаты, свидетельствующие о перспективности применения наноразмерного магнетита для выделения тетрациклинов. В научной группе проводились работы по сочетанию магнитной твердофазной экстракции со спектрофотометрическим определением лекарственных веществ.
