Аннотация:Резюме (рус)Воспалительные заболевания кишечника (ВЗК) — это группа заболеваний, развитию которых способствует широкий ряд факторов, а существующее лечение направлено на купирование симптомов. Характерной чертой ВЗК является воспаление слизистой оболочки кишечникаи последующее возникновение воспалительных процессов, что приводит к нарушению пищеварения. Нарушение слизистого барьера приводит к развитию патологий, в результате активируется локально иммунная система, и развивается воспаление. В ответ на воспаление, проникающее в ткань кишечника, макрофаги поляризуются по провоспалительному типу, секретируяпровоспалительные цитокины. В результате развивается хроническое воспаление, со временемспособствующее развитию осложнений, в том числе опухолей. Схожий патогенез развития ВЗКнаблюдается у животной модели ВЗК — мышей с нокаутом гена муцина 2. У мышей Muc2-/- истощен слизистый слой толстого кишечника, в норме формируемый белком муцина 2. Отсутствиенормального слизистого барьера приводит к развитию хронического воспаления, по симптоматике и проявлениям схожего с таковым у пациентов с ВЗК. Немаловажной характеристикой данноймодели ВЗК является возможность изучать широкий спектр симптомов на одной модели без влияния искусственных индукторов. Такие особенности мышей с нокаутом гена муцина 2 делают этулинию мышей не только эффективной моделью ВЗК, но и моделью хронических воспалительныхпроцессов в целом.Abstract (eng)Inflammatory bowel diseases (IBD) is a group of diseases, which development is promotedby a wide range of factors. The existing treatment is aimed at relieving symptoms. A characteristic feature of IBD is inflammation of the intestinal mucosa and the subsequent occurrence of inflammatoryprocesses, which leads to digestive disorders. Violation of the mucosal barrier leads to the development of pathologies, as a result, the immune system is activated locally, and inflammation develops.Macrophages penetrating into the intestinal tissue in response to inflammation, then they are polarizedin to proinflammatory type and start secreting proinflammatory cytokines. As a result, chronic inflammation develops, eventually contributing to the development of complications, including tumors. A similar pathogenesis of IBD development is observed in an animal model of IBD — mice with a knockoutof the mucin 2 gene. The mucosal layer of large intestine is depleted in Muc2-/- mice, normally formedby mucin 2 proteins. The absence of a normal mucosal barrier leads to the development of chronic inflammation, which symptoms and manifestations are similar to those in patients with IBD. An importantcharacteristic of this IBD model is the ability to study a wide range of IBD symptoms on one model, without the influence of artificial inducers. Such features of mice with a knockout of the mucin 2 gene make this line of mice not only an effective model of IBD, but also a model of chronic inflammatory processesin general.
