Аннотация:The content of intracellular adenosine-5′-triphosphate (ATP) is the main indicator of metabolic status of cells and their viability. Bioluminescent ATP-assay based on luciferin-firefly luciferase system enables rapid and specific measurement of ATP the detection limit of 10-13 M. The main feature of this method is the possibility to quantify slow-growing microorganisms such as Mycobacterium. Especially it is important for real-time monitoring of microbiological quality of such biotechnological product as BCG (Bacillus Calmette-Guerin) vaccine for tuberculosis disease for which the traditional plate count analysis takes four weeks.
We developed and validated a simplified ATP method to control the specific activity (CFU/mg) of the vaccine before and after freeze-drying [2]. Total time of the analysis is 30 min for liquid cell suspension and 90 min for lyophilized form. The method is characterized by good reproducibility, stability, and simplicity of processing the measurement
